Lipopolysaccharide Induced Activation of Toll Like Receptor 4 in Isolated Rat Heart Suggests a Local Immune Response in Myocardium

چکیده مقاله:
Background: Myocardial dysfunction is one of the major complications in patients with sepsis where there is a relationship between the blood level of cytokines and the onset of myocardial depression. In many cases of sepsis, the presence of Lipopolysaccharide (LPS) has been established. LPS Binding Protein (LBP) bound endotoxin is recognized by CD14/toll-like receptor-4 (TLR4) complexes in innate immune cells which stimulates TNF-α release. Objectives: To investigate whether isolated rat heart is capable of producing TNF-α locally through TLR4 activation by LPS. Methods: Using langendorff method, LPS in 120 mL of the modified KrebsHenseleit buffer solution (KHBS) at final concentration of 1 µg/mL was perfused at recycling mode. The effect of LPS on cardiac function was evaluated. To assess the TLR4 activity and TNF-α release, western blotting, real time-PCR, and ELISA were used. Results: Compared with control, coronary perfusion pressure (CPP) as well as left ventricular developed pressure (LVDP), maximum and minimum rates of the left ventricular developed pressure (dP/dtmax; dP/dtmin; p

متن کاملtoll-like receptor 4 in ventilator-induced lung injuries
toll like receptors (tlrs) recognize pathogens and generate an immediate defense response by inducing the production of pro-inflammatory cytokines, which rapidly destroy or limit the pathogens. in their bridging role, tlr downstream signals link innate and adaptive immunity, particularly by mediating dc maturation and activation of pathogen specific t lymphocytes. these pathways lead to the act...
متن کاملmetformin alleviates lipopolysaccharide-induced acute lung injury through suppressing toll-like receptor 4 signaling
signaling amp-activated protein kinase (ampk), an energy sensing enzyme, has been implicated in controlling inflammation. in this study we investigated whether activation of ampk by metformin could protect the lung from lipopolysaccharide (lps)-induced acute injury by inhibitingng tlr4 pathway. male wistar rats were randomly divided into 3 groups (n=6): control group received normal saline (0.5...
متن کاملSignal transduction by the lipopolysaccharide receptor, Toll-like receptor-4.
An understanding of lipopolysaccharide (LPS) signal transduction is a key goal in the effort to provide a molecular basis for the lethal effect of LPS during septic shock and point the way to novel therapies. Rapid progress in this field during the last 6 years has resulted in the discovery of not only the receptor for LPS - Toll-like receptor 4 (TLR4) - but also in a better appreciation of the...
متن کاملIn silico Study of Toll-Like Receptor 4 Binding Site of FimH from Uropathogenic Escherichia coli
Introduction : The innate immune system as the first line of defense against the pathogens recognizes pathogen-associated molecular patterns (PAMPs) by Toll-Like Receptors (TLRs). Interaction of bacterial PAMPs by TLRs results in activation of innate and acquired immunity. FimH adhesin, a minor component of type 1 fimbriae encoded by Uropathogenic Escherichia coli (UPEC) is a PAMP of TLR4 tha...
متن کاملPrenatal zinc supplementation ameliorates hippocampal astrocytes activation and inflammatory cytokines expression induced by lipopolysaccharide in a rat model of maternal immune activation
Objective: There is evidence that gestational exposure to lipopolysaccharide (LPS) results in fetal zinc deficiency, and eventually neurodevelopmental abnormalities. In this study, we utilized a rat model of maternal immune activation (MIA) to investigate the possible neuroprotective effect of zinc supplementation throughout pregnancy on hippocampal astrocytes activation as well as inflammatory...
